Metabolomics is the science of studying endogenous metabolites, and its research strategies mainly include non-targeted and targeted. Untargeted metabolomics generally uses high-resolution mass spectrometry to obtain rich metabolite information but has disadvantages such as complex data, poor reproducibility, and narrow linear range. Targeted metabolomics generally uses triple quadrupole mass spectrometry, which has greatly improved data quality, but usually only detects known metabolites with low coverage. However, semi-targeted metabolomics combines the advantages of both, and its principle is mainly to obtain ion pairs of metabolites in the sample to be analyzed. This strategy can achieve a good compromise between efforts in metabolite coverage, reliability, method construction, and data processing.

Service Content of Semi-Targeted Profiling of Metabolites

For this service, Creative Proteomics typically performs semi-targeted analysis of metabolites using a UPLC-MS system consisting of a photodiode array detector (PDA), a mass spectrometer equipped with Masslynx-controlled electrospray ionization (ESI) source, and a chromatograph. First, we perform chromatographic separation of the analytes using a chromatographic column, followed by MS detection of the samples. For relative quantification, UPLC-MS analysis is performed in selected ion monitoring (SIM) mode. Finally, the assay data are analyzed and compared by bioanalytical software.

Requirements for Samples

  • Microbial and cellular samples require rapid blunting of metabolic activity (quenching) while keeping cells from lysing.
  • Animal body fluids (e.g., urine, blood, tissues, organs, saliva) should be rapidly pretreated after sampling, e.g., by adding anticoagulants, and preservatives, and immediately stored at -80℃.
  • Plant samples are frozen (liquid nitrogen) rapidly after collection and then transferred to -80℃ for storage.
  • Serum samples should not be less than 200 µL and avoid repeated freezing and thawing.
  • Urine samples 1 mL, sent frozen at -80℃.
  • Please provide the specific concentration, volume, preparation time, and source of each sample. And inform the sample information as well as the control and experimental samples (if there is a group, describe the group information in detail.)
