Absolute quantitative analysis is a mass spectrometry-based targeted quantitative proteomics technique widely used in various quantitative proteomics studies. The rationale for this technique is to use stable isotope-labeled synthetic peptides as internal standards to differentiate natural peptides. Quantitative techniques are then used to obtain mass spectrometry ratios of internal standard peptides to native peptides, enabling precise and quantitative determination of absolute levels of proteins and post-translationally modified proteins. Simple to operate and sensitive to results, AQUA assay technology has become an effective method to measure protein levels and post-translational modifications directly from cell lysates.
Service Content of Absolute Quantitative Analysis (AQUA)
In the absolute quantitative analysis service, Creative Proteomics uses a stable isotope-labeled synthetic peptide as an internal standard, which is introduced into cell lysates at known concentrations during digestion. The universal signal response factor was the same for all assayed proteins. During the detection process of the tandem mass spectrometer, the hydrolyzed protein samples were analyzed by the SRM method, and the natural peptides and isotope-labeled AQUA internal standard peptides could be directly detected and quantified. In this service, we can not only provide mass spectrometry results, but also provide a series of downstream service solutions such as GO functional annotation and enrichment analysis, differentially expressed protein cluster analysis, protein interaction network analysis, and differential protein statistical analysis.

Requirements for Samples
- The animal tissue samples should be > 200 mg.
- The number of cell samples should be > 107.
- The microbial (bacteria/fungus) sample dry weight should be > 200 mg.
- The protein sample should be > 50 μg and the concentration is greater than 80%.
- Please note that all samples should be stored at -80°C, do not freeze and thaw samples repeatedly, and use sufficient dry ice for transportation to reduce the possibility of sample degradation during transportation.
- Please provide the specific concentration, volume, preparation time, and source of each sample. And inform the sample information as well as the control and experimental samples (if there is a group, describe the group information in detail.)
