Creative Proteomics provides a high-quality, rapid protein phosphorylation identification service to support those involved in protein phosphorylation research.
Phosphorylation of proteins is simple, flexible, and reversible, and is very common in eukaryotes. Protein phosphorylation is a reversible process catalyzed by enzymes, mainly connecting phosphate groups to proteins, which makes the protein structure more complex, function more perfect, regulation more refined, and action more specific. Phosphorylation and dephosphorylation of proteins regulate almost all processes including cell proliferation, development, differentiation, cytoskeleton regulation, apoptosis, neural activity, muscle contraction, metabolism, and other life activities.
Service Content of Protein Phosphorylation Identification
Due to the low content and wide dynamic range of phosphorylated proteins in biological samples, before analysis of phosphorylated peptide samples using quantitative proteomics analysis methods, it is necessary to enrich the modification to improve the abundance of modified proteins. Creative Proteomics provides customers with one-stop service from protein extraction, proteolytic digestion, phosphorylated peptide enrichment, peptide separation, mass spectrometry analysis, mass spectrometry raw data analysis to bioinformatics analysis. In the process of providing services, we firstly enrich the phosphorylated peptides efficiently, and in the process of enriching the peptides, separate the monophosphorylated peptides and the polyphosphorylated peptides step by step, to perform mass spectrometry separately. The analysis avoids the influence of polyphosphorylated peptides on the identification and quantification of monophosphorylated peptides due to the problem of ionization efficiency during mass spectrometry analysis.
Mass Spectrometry Facilities and Projects
- Orbitrap Fusion Mass Spec Platform
- LC-20AT, SHIMADZU High Performance Liquid Chromatography
- Thermo Scientific™ Q Exactive™ Hybrid Quadrupole-Orbitrap Mass Spectrometer
Requirements for Samples
- Samples provided by customers can be protein samples or cell samples.
- Protein sample: total protein > 800 μg, protein purity > 90%.
- Cell sample: The sample is recommended to contain no less than 5 × 107 cells.
- Note, try to avoid trypsin when collecting cells. Phosphatase inhibitors are required throughout sample processing and collection. Please store all samples at -80℃, do not freeze and thaw samples repeatedly, and transport protein samples with sufficient dry ice.
- Please provide the specific concentration, volume, preparation time, and source of each sample. And inform the sample information as well as the control and experimental samples (if there is a group, describe the group information in detail.)
