The basic principle of multidimensional protein identification (MudPIT) is to use an enzyme (Trypsin) to enzymatically dissociate a protein solution or a complex mixture in SDS-PAGE bands into a mixture of peptide fragments, and then perform multidimensional capillary liquid chromatography separation and tandem mass spectrometry analysis of the peptide mixture, and finally use the corresponding database to search and match, which can identify hundreds of proteins at the same time. Compared with traditional two-dimensional electrophoresis technology, multidimensional protein identification has higher sensitivity and a more comprehensive detection range. Whether it is a whole tissue, body fluid, or its extract, multi-dimensional protein identification technology can identify protein molecules or peptide molecules. Service Content of Multi-Dimensional Protein Identification

Our multidimensional protein identification service combines cation-exchange pre-fractionation of tryptic peptides and RP HPLC separation to analyze the entire proteome of cell- or tissue-type protein extracts. Double-enzymatic digestion was used to increase the number of observed peptides, and the samples were then subjected to LC-MS/MS and protein identification using a mass spectrometry instrument.

Requirements for Samples

  • Customer-provided samples can be either strips from SDS-PAGE gels or solution samples. You will need to send us the stained gel image and the protein blot image.
  • The total amount of protein is 50-200 μg, the protein purity is > 90%, and the protein buffer does not contain detergent NP40, Triton X-100, etc.
  • Note that samples must be shipped with sufficient dry ice.
  • Please provide the specific concentration, volume, preparation time, and source of each sample. And inform the sample information as well as the control and experimental samples (if there is a group, describe the group information in detail.)
